任豆叶片的愈伤组织诱导和植株再生

摘 要:

为建立任豆(Zenia insignis)无菌苗培养体系, 以种子作为实验材料, 采用不同消毒方法对其灭菌效果进行比较。并以无菌苗叶片为外植体, 以MS为基本培养基, 研究不同配比的植物生长调节剂对愈伤组织诱导、不定芽诱导和生根培养的影响。结果表明: 种子用0.1%升汞消毒10 min, 无菌苗获得率最高。诱导愈伤组织的最佳培养基配比为MS+2 mg·L-1 6-BA+0.01 mg·L-1 IBA+0.01 mg·L-1 2,4-D, 诱导率为82.88%。6-BA在不定芽诱导中起着关键作用, 诱导不定芽最佳培养基为MS+0.01 mg·L-1 IBA+3.2 mg·L-1 6-BA。生长素对任豆的生根有重要影响, MS+0.1 mg·L-1 NAA为生根培养基可获得98.23%的生根率。

Callus induction and plant regeneration from the leaf of Zenia insignis

Abstract:

In order to establish the regeneration system of Zenia insignis, the seeds were selected as experimental materials, and the sterilization effects were compared by different disinfection methods. The effects of different ratio of plant growth regulators on callus induction, adventitious bud induction and rooting culture were studied with leaves as explants and MS as media. The results showed that seeds with 1% mercuric chloride for 10 min, obtained the highest rate of aseptic seedling. The optimum medium proportion of callus induction was MS+2 mg·L-1 6-BA+0.01 mg·L-1 IBA+0.01 mg·L-1 2,4-D, and the induction rate was 82.88%. 6-BA plays a key role in the adventitious bud induction and the best medium for adventitious bud induction was MS+0.01 mg·L-1 IBA+3.2 mg·L-1 6-BA. Auxin has an important effect on the rooting, the medium of MS+0.1 mg·L-1 NAA for rooting could get the rooting rate of 98.23%.
